Absolute Quantification
Absolute quantification by qPCR offers an accurate way to determine the precise copy number of a target nucleic acid molecule within a sample.This approach enables the accurate quantification of a target molecule’s copy number within samples. By conducting qPCR on a dilution series of a sample with a known amount of the desired target molecule, users can generate a standard curve of Ct values corresponding to a given amount of the target. The Ct values of the samples under examination are then compared to this reference curve and a precise estimate of the target molecule’s abundance in each unknown sample calculated. This is the method of choice for many diagnostic applications and for analysis of species, allele, and transcript splice-variant abundance in a wide range of biological samples.
