High Fidelity PCR
High Fidelity PCRFor some applications such as cloning and site directed mutagenesis, or when preparing a template for sequencing, the number of mutations introduced during PCR needs to be kept to a minimum.
DNA polymerases occasionally incorporate mismatched bases into the extending strand, however high fidelity PCR makes use of DNA polymerases that possess a 3’-5’ exonuclease or ‘proofreading’ activity. This activity is used to excise incorrectly incorporated nucleotides, which are then replaced with the correct nucleotides, allowing faithful replication of the target DNA of interest.
PCR Biosystems offer a range of versatile and robust high fidelity enzymes engineered for all PCR applications where greater sequence accuracy is required, including applications with challenging reaction conditions such as long-range PCR, fast PCR or the amplification of GC-rich targets.
