High Fidelity PCR

For some applications such as cloning and site directed mutagenesis, or when preparing a template for sequencing, the number of mutations introduced during PCR needs to be kept to a minimum.

DNA polymerases occasionally incorporate mismatched bases into the extending strand, however high fidelity PCR makes use of DNA polymerases that possess a 3’-5’ exonuclease or ‘proofreading’ activity. This activity is used to excise incorrectly incorporated nucleotides, which are then replaced with the correct nucleotides, allowing faithful replication of the target DNA of interest.
