How much magnesium chloride should I add to my PCR or qPCR reaction?
Further addition of PCR enhancers or MgCl2to the reaction is not recommended, as the buffer’s composition has been optimised to maximise PCR success rates. However, if developing a customised assay, additional MgCl2 should be added in 0.5 mM increments up to a maximum of 5-7 mM and results compared to unmodified reactions.
How much ROX should I add to my qPCR reaction?
The dye is supplied at a 50 μM concentration that can be used as a 100x stock solution for instruments requiring high ROX concentration (500 nM) and as a 1000x stock for instruments requiring a low ROX concentration (50 nM). Use our qPCR Selection Tool to identify the appropriate ROX concentration for your instrument. If your instrument model is not listed, please refer to the manufacturer’s instructions or contact our technical support team.

Should I add extra magnesium chloride to my PCRBIO products?
PCR Biosystems polymerase kits and mixes are supplied with an optimised magnesium chloride concentration and generally do not require any further addition. However, some assays may require further customisation.
