DNA methylation is a heritable epigenetic mark involving the addition of a methyl group to the 5′ position of cytosine by DNA methyltransferases. It plays an important role in normal human development and is linked to a number of key processes including gene expression, aging, and carcinogenesis

PCR-based techniques are routinely used to study DNA methylation following the treatment of DNA with sodium bisulphite. This treatment involves the deamination of cytosines to uracil which are then converted to thymine in subsequent PCR amplification. 5-methylcytosine remains intact during treatment and is amplified as cytosine, thereby allowing methylated cytosines to be distinguished from unmethylated cytosines when the PCR products are analysed. PCR Biosystems provides a range of reagents to support DNA methylation analysis using PCR, real-time PCR and high resolution melt (HRM) analysis.
